Elizabeth came to New England on the "Anne" in 1623, accompanied by her daughters. Elizabeth was rated in the Plymouth tax list of 1632/33, and was one of the first purchasers of Dartmouth.

The Early Records of Plymouth reveal the fact that Elizabeth was a woman, not only of social position in the community, but one of force. The term "Mistress" Elizabeth Warren that is shown in the records is a designation that was not common. She also lived without marrying for the next forty-five years, something that was very unusual for her time.

On 5 July 1635, Elizabeth Warren's servant, Thomas Williams, was accused of "speaking profane and blasphemous speeches against ye majestie of God," and "There being some dissention between him and his dame, she after other things, exhorted him to fear God & do his duty" by the General Court.

Elizabeth died at Plymouth, at the age of ninety years. Emily Warren gives the date of death as 2 Oct 1673 and 22 Oct 1673 as her burial date according the the Plymouth records. Other authors have questioned that this might be a mistake. They give her death as 22 Oct 1673 and her burial as 24 Oct 1673.

"Mistris Elizabeth Warren an aged widdow aged above 90 years Deceased on the second of October 1673 whoe haveing lived a Godly life Cam to her Grave as a shok of Corn fully Ripe shee was honoralbey buried on the 24th of October aforesaid."
